From its early history as a North American working dog used to retrieve fishing nets and perform human water rescues, the beautiful and intelligent Newfoundland dog has carved out a well-deserved place in the heart of dog lovers around the world. Their characteristic large size, marked by heavy bones, powerful musculature, webbed feet, and thick hair coat make the Newfoundland particularly adept at tasks involving swimming. However, they are just as capable and content pulling carts on land and performing other land-based tasks. In addition, their generally calm, loyal, and affable temperament have helped establish them as great family dogs.  

Over the years, Newfoundland breeders have selectively bred dogs that have displayed the most desirable characteristics in an effort to improve their breed. Unfortunately, alongside these desirable traits, sometimes the predisposition to produce offspring with certain inherited diseases are also silently passed from generation to generation in the form of genetic mutations. Prevention Is Smart Breeding- Urate Bladder Stones in the Spanish Water Dog

Though reliable accounts marking the early origins of the Spanish water dog (SWD) are lost to history, likely ancestors of the SWD (and probably the Portuguese water dog) were described in the Iberian Peninsula around the 12th century. Primarily used as a herding breed, the Spanish water dog has also been trained to perform tasks for fishermen and hunters including towing boats to shore, gathering fishing nets, and retrieving game from waterways. Though still used as working dogs in modern day Spain and other countries, the demand for Spanish water dogs dropped precipitously during the industrial revolution of the late 18th and early 19th centuries when machines took over many tasks historically performed by the SWD and other breeds. This decrease in demand led to a decline in the world Spanish water dog population which still remains relatively small today.

In general, dog breeds with smaller population sizes tend to also have a smaller amount of genetic diversity within the breed. Preventing Inherited Urate Bladder Stones in the Bulldog

Though the cruel bull-baiting practices for which the ancestors of bulldogs were used have long passed, the enthusiasm for the modern bulldog as a friendly companion animal is as fervent and robust as ever. Ranked the 4th most registered dog in the AKC, it’s no secret to most that the bulldog has established itself as one of America’s favorite canines. Through selective breeding practices, the previously fierce and aggressive bulldog of old has given way to a gentle, even tempered, yet strong willed breed with copious amounts of character; in both temperament and physical attributes. Unfortunately, like other purebred dogs, during the course of breed development, some inherited genetic diseases have come to light that can make life difficult for bulldogs and those that love them. Luckily, through the use of genetic testing technologies and selective breeding, some of these inherited diseases can be completely eliminated (or greatly reduced in frequency) from blood lines. One example of an inherited disease that can now be prevented in the bulldog is an inherited form of urinary stones known as hyperuricosuria (HUU), caused by a genetic mutation in the SLC2A9 gene.

In this fourth and final blog in a series about preventable inherited diseases in the Labrador retriever (see previous blogs here; part one, part two, and part three), we will examine a potentially life-threating urinary disease and a cause of blindness in dozens of dog breeds including the Labrador.

Cystinuria

Urinary tract obstruction is one of the most urgent and potentially life-threatening conditions in dogs. One common cause of urinary obstructions in dogs is the presence of bladder stones which leave the bladder during urination and become lodged in the urethra. The inability to urinate results in a toxic buildup of certain electrolytes and waste products in the body including potassium, which tends to be the most concerning in regards to health effects. Excess potassium in the body prevents the heart from beating normally, ultimately resulting in death due to cardiac arrest. Though it is not the only cause of bladder stones in Labradors, a genetic mutation in the SLC3A1 gene is reported to cause an early-onset disease known as cystinuria, which can result in bladder stone formation.

Are Dalmatians the only breed to develop urate bladder stones?

It is no secret among many Dalmatian owners and their breeders that their beautiful breed is prone to developing bladder stones (uroliths) made of urates as they have historically been the poster puppies for the inherited condition, known as hyperuricosuria. However, many don’t realize that the same genetic mutation responsible for these stones has been reported in over a dozen breeds of dog, although at a much lower frequency.

Urates are formed from uric acid, a product of the breakdown of natural compounds of the body and from our food, known as purines. Affected dogs are predisposed to the formation of these bladder stones due to a mutation in the SLC2A9 gene1; a gene that serves as the blueprint for a protein responsible for transporting uric acid in the body. In dogs free of this mutation, this transporter plays an important role in transporting uric acid from the blood into the liver for degradation and from urine produced in the kidney, back into the blood. Affected dogs cannot perform these tasks adequately, thus resulting in a high level of uric acid in both the blood (hyperuricemia) and urine (hyperuricosuria).
